Loss of PGC1α drives extracellular matrix remodelling in prostate cancer through CTHRC1. (PubMed, bioRxiv)
Importantly, recombinant CTHRC1 rescues adhesion defects, indicating that its extracellular pool mediates this phenotype, whereas deglycosylation abolishes its pro-adhesive function, revealing a mechanistic requirement for glycosylation. Collectively, our findings uncover an early, cell-intrinsic ECM remodelling program driven by PGC1α loss and identify the PGC1α-CTHRC1 axis as a mechanistic and clinically relevant regulator of PCa aggressiveness.

Genomic profile of an IDH-wild-type glioblastoma diagnosed following TNF-α inhibitor therapy: a molecular case study. (PubMed, Front Oncol)
TNF α inhibitors such as adalimumab are widely used for autoimmune diseases, yet their long-term impact on tumor development in genetically susceptible individuals remains incompletely defined...While no causal inference can be made, this analysis identifies both canonical and atypical genomic alterations in a GBM arising after prolonged TNF-α targeted therapy. This temporal relationship provides a basis for studying possible convergence between TNF-α signaling and GBM-associated pathways, and underscores the importance of genomic risk stratification when considering TNF-α inhibitor therapy.

Loss of Kmt2c/d promotes gastric cancer and confers vulnerability to mTORC1 and PD1 inhibition. (PubMed, J Clin Invest)
Combination of mTORC1 inhibition and anti-PD1 immunotherapy markedly suppressed tumor growth in immune-competent mice. Together, these findings reveal the role of Kmt2c/d loss in gastric cancer initiation and suggest the potential therapeutic strategies for KMT2C/D-deficient gastric cancer.

BRaf-mutant aNSPCs display increased Sox2 protein, but not mRNA levels, suggesting post-transcriptional regulation. Sox2 phosphorylation at T118 is promoted by BRafV600E, potentially stabilizing the protein during aNSPC transformation, although the involvement of downstream kinases cannot be excluded.

Biobank of genetically defined murine prostate cancer tumoroids uncovers oncogenic pathways and drug vulnerabilities driven by PTEN-loss. (PubMed, Cell Rep Methods)
Notably, these compounds also inhibited the growth of several human PCa cell lines and displayed synergistic effects when combined with the standard-of-care antiandrogen enzalutamide. Together, our findings provide evidence that murine tumoroids are versatile preclinical models for studying PCa tumorigenesis and drug sensitivities to develop therapeutic options for PCa patients.

Deficiency of PTEN Confers Hypersensitivity to Fatty Acid-Mediated ER Stress in Transformed Hepatocytes. (PubMed, Int J Mol Sci)
Xenograft studies showed that PTEN KO HCC tumors progressed faster than did the control tumors when mice were fed with normal chow and slower under a high-fat diet. We suggest that while the health risks of a fatty acid-rich diet to liver function and the increased propensity to develop HCC are prominent, once a PTEN-deficient HCC has been established, it exposes vulnerability to lipid overload that can be exploited through diet and pharmacological interventions.
