Experimental study of [68Ga]Ga-HER2-RGD, a dual-target molecular probe, for breast cancer PET/CT imaging. (PubMed, EJNMMI Res)
[68Ga]Ga-HER2-RGD demonstrates specific dual-receptor-targeting properties towards HER2 and integrin αvβ3, as evidenced by in vitro and in vivo studies. This radiopeptide shows promise as a novel PET agent for the noninvasive visualization of HER2 and/or integrin αvβ3 expression in breast cancer, and is capable of semi-quantitatively evaluating the expression levels of HER2 and/or integrin αvβ3 to a certain extent.

Therapeutic response to neoadjuvant chemotherapy in HER2-positive pure mucinous breast cancer: a case report. (PubMed, Int J Surg Case Rep)
She received six cycles of neoadjuvant docetaxel, carboplatin, trastuzumab, and pertuzumab. Persistent radiologic abnormalities may represent residual mucin rather than viable tumor cells, underscoring a potential limitation of imaging-based response evaluation. Recent Advances in the Treatment of HER2-Altered Solid Tumors (PubMed, Gan To Kagaku Ryoho)
Antibody-drug conjugates (ADCs), led by trastuzumab deruxtecan, have established clinically meaningful activity across multiple solid tumors in HER2 IHC 3+ populations and have further expanded into HER2-low and ultralow expression settings, supporting the concept of HER2 as a biological continuum rather than a binary biomarker...In gastroesophageal adenocarcinoma, treatment options beyond trastuzumab-based regimens continue to diversify: ADC-based standards in later lines are being complemented by next-generation HER2 antibodies, inclu ding recent phase Ⅲ evidence supporting anbenitamab after prior trastuzumab exposure, which may further inform sequencing decisions. Collectively, these advances highlight an emerging precision model in which therapeutic selection and rational sequencing are guided by integrated assessment of HER2 protein expression, ERBB2 genomic status, prior HER2-directed exposure, and toxicity risk (notably ADC-associated interstitial lung disease). Standardization of high-fidelity diagnostic workflows-spanning IHC/ISH quality assurance, re-biopsy when feasible, and context-appropriate use of circulating tumor DNA-will be essential to ensure consistent patient identification and to enable optimal deployment of ADCs, TKIs, and antibody-based therapies across histologic boundaries.

Chelator-Free Radiometal Labeling Inside Engineered Affibodies. (PubMed, Angew Chem Int Ed Engl)
All retain their metal cargo when challenged with cellular concentrations of glutathione, while only the bismuth-affibody complex withstands a challenge with 100 equivalents of strong chelators, even over two weeks. We demonstrate that, a bismuth‑loaded affibody retains binding affinity to the HER2 receptor comparable to the wildtype affibody, while selectively binding and retaining 213Bi, a promising radioisotope for targeted alpha therapy, thereby enabling targeting of HER2‑overexpressing SKBR3 cancer cells.

Antibody-drug conjugate therapy in a renal transplant patient with bladder cancer: A case report. (PubMed, Clin Nephrol)
After initial treatment with transurethral resection of the bladder tumor (TURBT), the patient underwent regular bladder instillations of epirubicin...Due to the patient's renal impairment (baseline creatinine 107.8 µmol/L, glomerular filtration rate (GFR) 79.19 mL/min/1.73m2) and human epidermal growth factor receptor 2 (HER2) overexpression (2+), the decision was made to employ an antibody-drug conjugate (ADC) targeting HER2 (disitamab vedotin, 120 mg intravenously). The report outlines the patient's background, the progression of his disease, the decision-making process behind the use of ADC therapy, and the subsequent response to treatment. The successful control of the disease with ADC therapy in this complex patient population highlights its potential as an effective and less toxic alternative to conventional chemotherapy, particularly in patients with recurrent bladder cancer after renal transplantation.

Prognostic impact of p53 and HER2 immunohistochemistry profiles in colorectal carcinoma. (PubMed, Pathol Oncol Res)
HER2 expression was infrequent and showed no prognostic relevance; however, HER2-positive tumors were significantly associated with CK7 expression, supporting an "extraintestinal" immunophenotypic profile reminiscent of right-sided or non-intestinal differentiation. In conclusion, while p53 and HER2 expression lacked independent prognostic value, their distinct associations with tumor phenotype underscore biologically relevant CRC subgroups.
